4X Upper Gel Buffer contains Tris and SDS at the appropriate pH and concentration for convenient and quick preparation of polyacrylamide gels for protein analysis by SDS-PAGE. Dilution of the Stacking Gel 4X buffer (0.5M Tris pH 6.8 and 0.4% SDS) to a 1X solution results in the standard, 0.125M Tris, 0.1% SDS concentrations routinely used in the preparation of the stacking gel for SDS-PAGE.
