Products Description

源自嗜熱細菌的 Ampligase® Thermostable DNA Ligase 是具NAD+依賴性的熱穩定性連接酶 (thermostable ligase),相較傳統 DNA 連接酶於高溫下更穩定且具活性。 其半衰期在 65°C 下為 48 小時,在 95°C 下 49 小時。已證實 Ampligase DNA Ligase 至少在 500 個熱循環 (94 °C/ 80 °C) 或 16 小時的循環中仍具有活性。
Ampligase DNA Ligase 對連接 blunt-ended DNA 以及 RNA 或 RNA:DNA hybrids 沒有檢測活性。

Reaction Buffer: 10X Ampligase Reaction Buffer contains 200 mM Tris-HCl (pH 8.3), 250 mM KCl, 100 mM MgCl2, 5 mM NAD, and 0.1% Triton® X-100.

Unit Definition: One unit of Ampligase Thermostable DNA Ligase catalyzes the ligation of 50% of the cos sites present in 1 μg of bacteriophage lambda DNA in 1 minute at 45°C.
*Note: One Ampligase unit is equivalent to at least 15 “cohesive end units” or the “nick closing units” of thermostable ligase described elsewhere.
